In the autumn of 1872, fourteen sisters from Gurtweil arrived at the Novitiate in Belle Prairie. To accommodate them, twenty sisters moved to St. Louis, where Herman Leygraff, the pastor of St. Agatha's, offered a home until a new motherhouse could be built.
By 1873, the Kulturkampf had intensified, forcing the entire community to leave Germany. Some sisters went to Rome, while others settled in Bosnia and Hungary. In September, Muehleisen met forty-nine sisters in New York and guided them to St. Louis, where the new Motherhouse in O'Fallon, Missouri, was completed by July 1875.
The community in O'Fallon reorganized itself with a new rule and constitution, and in 1878, it was incorporated under Missouri law as St. Mary's Institute of O'Fallon. Augusta Volk is recognized as the founder of the congregation. A new novitiate was established in O'Fallon. The Sisters of O'Fallon became a congregation of pontifical rights in 1918 and adopt the post-nominal CPPS. As of 2019, about 100 sisters served in the O'Fallon congregation.
